Abstract
Objective To explore the correlation of Toll-like receptor 4 (TLR4) promoter region gene polymorphism and environmental factors and unexplained recurrent abortion in female of Xinjiang Uygur Autonomous Region.Method 40 cases of unexplained recurrent spontaneous abortion female from January 2015 to June 2016 were enrolled in case group. Meanwhile, 40 cases of normal early pregnancy female in the same period were enrolled in control group. Female in the two groups were investigated by basic situation questionnaire, peripheral blood DNA extraction and sequencing. Meanwhile, Western blotting and real-time fluorescence quantitative polymerase chain reaction were used to detect TLR4 mRNA and protein expression in decidua tissues.Result Female who≥35 years old and BMI≥24 kg/m2 of case group were more than control group (P<0.05). In the TLR4-ss77136219 gene locus, the frequency of G carrying allele in case group was significantly higher than that in control group (P<0.05). The positive rate of TLR4 expression in decidua tissue of case group was significantly higher than that in control group (P<0.05).On the TLR4-ss77136219 gene locus, the expression level of TLR4 mRNA and TLR4 protein in GG genotype female was significantly higher than that of AA genotype female (P<0.05).Conclusion The genetic variation of TLR4 promoter region (A>G), age and overweight are closely related to the incidence of unexplained recurrent spontaneous abortion in female of Xinjiang Uygur Autonomous Region.关键词
