P53基因在大鼠唾液腺放射损伤中的表达变化及意义
Expression of P53 in salivary glands of rats after irradiation

Abstract
Objective:To investigate the expression of P53 in salivary glands of rats after 60Coγ-ray irradiation.Methods:Forty-eight SD rats were randomly divided into a control group and an experimental groups (n=24 per group).60Coγ-ray irradiation was performed once at a dose of 18 Gy on head and neck region of experimental rats.All rats were weighted once a week.The histopathological changes of salivary glands were observed by hematoxylin-eosin (HE) staining.The mRNA and protein expressions of P53 in rats salivary glands were determined by qPCR and immunohistochemistry,respectively.Results:The body weight of the experimental rats was decreased temporarily at 2 weeks post-irradiation,and then gradually increased.The body weight in the experimental group was notably lower than that in the control group within 12 weeks of irradiation (P<0.05).Morphological changes of the radiated gland cells showed interstitial vascular congestion.The number of vacuoles in the salivary gland acinar cells increased significantly.The mRNA and protein expressions of P53 in the experimental group were gradually elevated at 1,4,and 8 weeks after irradiation (P<0.05),but no significant difference during 8 to 12 weeks.The expression of P53 was markedly higher in the experimental group than that in the control group at any time point of irradiation (P< 0.05).Conclusion:The expression of P53 was up-regulated by radiation in rats and was significantly increased along with the time of irradiation.The P53 gene might be closely related to radiation-induced salivary gland injury.关键词
