摘要

Abstract

Thrombospondin1(TSP-1)gene plays a variety of biological functions in model animals.Tran-scriptome analysis suggests that TSP-1 may play an important role in the development of sheep follicles.To ex-plore the sequence structure and biological function of TSP-1 gene,the full-length of TSP-1 gene of Aletai sheep CDS region was amplified by PCR,the protein sequences and functional region of the TSP-1 gene were analyzed by software,and using the method of fluorescence quantitative detection of PCR the organization ex-pression of 5 1-year-old Aletai sheep was tested.The results showed that the TSP-1 gene was 3 519 bp in length,encoding 1 172 amino acids,and there were 2 nonsense mutations compared with the predicted se-quence.Phylogenetic tree analysis showed that the genetic distance between sheep and cattle and wild boar was close.TSP-1 protein structure was not stable,there was no transmembrane structure with a signal peptide,there may be 6 N-glycosylation and 50 potential glycosylation sites in O-loci,and 44 potential phosphorylation sites, binding sites and a N type Ga ion binding sites and TSP-3 protein containing 2 long repeats,3 short repeats and 6 C Ga ions.TSP-1 gene was expressed in 7 tissues of Aletai sheep,and the expression of TSP-1 gene in-creased gradually with the increase of follicle diameter.The results suggest that TSP-1 gene may play an impor-tant role in the development of ovine follicles.
