Expression of TCF7L2 and its relationship with platinum drug resistance in gastric cancer
摘要
Abstract
Objective To study the expression of TCF7L2 gene in gastric cancer and its relationship with plati-num resistance.Methods TCF7L2 mRNA of 15 gastric cancer tissues and adjacent cancer tissues were detected by qRT-PCR,and TCF7L2 protein expression of gastric cancer cell lines SGC-7901,HSC44,44AS3,N87,MKN45, MGC803,and immortalized normal gastric epithelial cells GES-1 were detected by western blot.The high TCF7L2 protein expression of gastric cancer cell line SGC-7901/TCF7L2 and idling control SGC-7901 cell line(SGC-7901/CON)by stable plasmid transfection technique,the resistance of cisplatin and carboplatin detected by cell counting.Results The expression level of TCF7L2 was significantly higher in the tumors than that in the non-carcinomatous gastric tissues.The expression levels of TCF7L2 in gastric cancer cell lines were also significantly higher than control cell line GES -1.The tolerance of resistant of SGC-7901/TCF7L2 cell line with high expression of TCF7L2 to cisplatin and carboplatin were better than than the control cell lines.Conclusion There is a positive relationship between expression of TCF 7L2 and occurrence and development of gastric carcinoma.It may be one of the reasons for its tolerance to platinum drugs.关键词
