Abstract

Objective:To establish a method by using target cell and UPLC/Q-TOF-MS for extraction the pharmacodynamic components in Chuanxiong Rhizoma.Methods:The human umbilical vein endothelial cells (HUVEC) were used as target cells,and the freeze-dried powder of Chuanxiong Rhizoma in logarithmic growth period was added.After 2 h incubation,the cells were collected and frozen rapidly with liquid nitrogen,the cells were broken by repeated freezing and thawing,and centrifuged to obtain a sample of cells after extraction.While compounds in the extract of cell samples were determined by UPLC/QTOF-MS,the effect of possible effective components that entered the cell of Chuanxiong Rhizoma was analyzed.Results:After HUVEC extraction two active ingredients ferulic acid and ligustilide were obtained.At the same time,the method was established for the determination of the two components.Conclusion:The analysis method of the target cell extraction and UPLC/QTOF-MS can be applied for screening out the active ingredients in Chuanxiong Rhizoma by specific binding effect between the compound and the target (receptor,channel,enzyme,etc.in the cell,so as to provide a new method for screening the active ingredients and drug mechanism study.
