丹参酮ⅡA联合伊马替尼对慢性髓细胞白血病K562细胞作用的实验研究
Effect of Tanshinone Ⅱ A Combined with Imatinib on K562 Cells of Chronic Myelocytic Leukemia
摘要
Abstract
Objective:To investigate the effect of Tanshinone Ⅱ A with Imatinib on the apoptosis of K562 cells and its related mechanisms.Methods:K562 cells were cultured and divided into six groups:normal K562 cells group,20 μM tanshinone ⅡA group,1 μM Imatinib group,and combined group(5 μM,10 μM,and 20 μM respectively plus 1 μM Imatinib).After treatment,the morphological changes of cells were observed under microscope;the cell apoptosis was detected by flow cytometry;and the mitochondrial activity was also observed by using JC-1 reagent and fluorescence microscope.Results:Tanshinone Ⅱ A promotes the cell apoptosis by inhibiting the mitochondrial transmembrane potential (P ＜ 0.05);and the inhibitory effects of tanshinone Ⅱ A were enhanced with the increase of its concentration.Conclusion:Tanshinone Ⅱ A can significantly enhance the apoptosis of K562 treated by Imatinib,which is related with mitochondrial pathway.关键词
