Abstract

Photo responsive sapogenin nanocapsules were prepared by a dual emulsification method to improve their stability and inhibiting activity on drug resistant bacteria. When soy lecithin, cholesterol and phaeophorbide are used as nanocapsule materials with a mass ratio of 50:10:1, the encapsulation efficiency, drug loading capacity, average particle size and zeta potential are 87.3%±4.6%, 13.5±1.2%, (155±5) nm and(-43.3±2.0) mV, respectively. The nanocapsule has a slow release effect without irradiation, but its drug release rate accelerates after irradiation at 650 nm. Its 50% inhibitory concentrations on drug resistant Escherichia coli and Staphylococcus aureus are 52.5 and 85.4 μg?mL-1,respectively,which decrease to 22.6 and 45.7 μg?mL-1respectively when combined with gentamycin (10 μg?mL-1). This result indicates that the nanocapsule can reduce and reverse bacterial resistance against gentamycin. Molecular docking results show that there are remarkable interactions between sapogenin and mannitol dehydrogenase (MDH) or Penicillin binding proteins (PBPs), which mean that the nanocapsules are involved in the inhibition of drug resistant bacteria.
