摘要

Abstract

In order to study the function of chitinase in Arthrobotrys oligospora in Xinjiang,the XJ-A1 chitinase AO-587 and AO-31 genes were amplified,cloned and sequencing.After sequencing,the gene and its encoded protein were taken to do bioinformatics and molecular feature analysis.The results showed that both chitinases belonged to the family of glycoside hydrolase 18 and contained the conserved sequence SXGG and VDGXDLDL in the chitinase catalytic region.AO-587 contains a signal peptide sequence,indicating that this protein belongs to the secretory protein.These two chitinase tertiary structures are similar,with a (α/β) 8 cylindrical structure.Phylogenetic analysis showed that the chitinase from different sources can be divided into three different evolutionary branches according to the difference of molecular weight,among which the isolation of chitinase AO-587 and AO-31 were closed with that of Drechslerells stenobrocha isolates,indicating that they may play similar roles in the process of infecting the host-nematode.
