Correlation of expression of protein inhibitor of activated signal transducer and activator of transcription 1 with tumor microenvironment of metastasisin gastric cancer and its significance in clinical prognosis

Abstract
Objective To investigate the correlation of expression of protein inhibitor of activated signal transducer and activator of transcription 1(PIAS1) with tumor microenvironment of metastasis (TMEM) of gastric cancer and its significance in clinical prognosis.Methods The expression of PIAS1,number of macrophagesand TMEM in 20 specimens each of superficial gastritis,atrophic gastritis,precancerous lesion,and 75 specimens of gastric cancer and adjacent normal tissue were detected immunohistochemically.TMEM was evaluated using anti-CD34 antibody as endothelial marker and anti-CD68 antibody as macrophage marker.The correlations between PIAS1 expression and number of macrophages,TMEM,clinical data including age,sex,stage,size and differentiation of tumor were analyzed.Results The positive expression rate of PIAS1 was lower and number of macrophages was higher in gastric cancer tissue than those in superficial gastritis,atrophic gastritis,precancerous lesion,adjacent normal tissues (26.7% vs 90.0%,80.0%,80.0%,48.0%,respectively),[(12.64 ±3.84) vs (7.45 ±2.28),(8.35 ±2.70),(8.95 ±2.56),(8.80 ±2.13)/high power field (HP),respectively],the differences were statistically significant (P<0.05).The number of macrophages and TMEM were higher in gastric cancer with metastasis than those without metastasis [(13.96±3.81) vs (11.80±3.66)/HP,(4.17±1.55) vs (3.26±1.32)].PIAS1 expression,number of macrophages and TMEM showed significant differences between different stages of gastric tissue (P<0.05).Number of macrophages and TMEM showed significant differences between those with positive and negative of PIAS1 expression (all P<0.05).Conclusions Lower-expression of PIAS1 was correlated with TMEM and prognosis of gastric cancer,which might be a prognosis marker and the therapeutic target of gastric cancer.关键词
