Follistatin-like 1 and inflammatory responses of diabetic atherosclerosis

Abstract
Objective To investigate the mechanism of the cytokine follistatin-like 1(FSTL1) in regulating macrophages to promote plaque formation in diabetic atherosclerosis.Methods To detect the expression level of FSTL1 in serum of patients with atherosclerosis and diabetic atherosclerosis by enzyme-linked immunosorbent assay(ELISA).A diabetic mouse model was established and detect the positive area of FSTL1 expression immunohistochemically.The levels of inflammatory cytokines in human macrophages were detected by ELISA after stimulating human macrophages by using oxidized low density lipoprotein (oxLDL) or/and FSTL1,and the mechanism of FSTL1 in the development of diabetic atherosclerosis was explored.Results The expression of cytokine FSTL1 was abnormally elevated in diabetic atherosclerosis than in atherosclerosis [(13.60±4.20) μg/L vs (9.90±3.50) μg/L (P<0.01)],and it was mainly located in macrophages.FSTL1 stimulated the release of inflammatory cytokines in macrophages and triggered local inflammatory responses.Conclusions The cytokine FSTL1 may be involved in plaque progression in diabetic atherosclerosis through activating inflammatory responses in macrophages.关键词
