摘要

Abstract

Objective:First micro oxidative injury model of vascular endothelial cells induced by oxidized low density lipoprotein in human brain was established,and then analyzed the model of HPS protection mechanism.Methods:Using oxidized low density lipoprotein (size 50g/ml) oxidation micro vascular endothelial cell injury model induced by the human brain,Radix Hedysari Radix Hedysari polysaccharide on the oxidative damage model,then used the MTT method to measure the protective effects of Radix Hedysari model,were used to detect the morphological changes of each group K1 vascular endothelial cells under the microscope.With Radix Hedysari (20g/ml) to interfere with brain micro oxidative injury model of vascular endothelial cells (time of day 24h),and then the extraction and purification of total cell protein (using cell lysate) gel electrophoresis after purification,cell protein,protein gel staining scanning spectra.Were randomly divided into controlled group (20cases),50g/ml oxidized low density lipoprotein in model group (20cases) and test group Hedysari polysaccharide drug intervention (20cases),extraction of each group of human brain microvascular endothelial cell total protein,protein map.The differential protein spots among each group were analyzed and identified by MS technique,and the differentially expressed protein spots were identified.Results:Radix Hedysari polysaccharide drug intervention experimental group cell survival rate,cell number,cell morphology than the human brain microvascular endothelial cells oxidative damage model group,after the separation and purification of human brain microvascular endothelial cell protein,established human brain microvascular endothelial cell protein electrophoresis and get protein;protein profiles obtained by the software of Hedysari polysaccharide drug intervention experiment group of human brain microvascular endothelial cell protein expression spectrum compared with the model group,the difference was statistically significant (P<0.05);analysis of the two group,there were 1 ldifferential protein spots,using the technology of MS identified 7different proteins.The identified proteins were identified this as heat shock protein 70and glutathione synthetase,net cavity calcium binding protein,ribosomal protein 40S and 78kDa Glucose regulated proteins,5of which were up-regulated and 2proteins down regulated.Conclusion:Radix Hedysari polysaccharide on oxidized low density lipoprotein induced oxidative brain micro vascular endothelial cell injury model has obvious protective effect,the mechanism is probably the main mechanism of Radix Hedysari polysaccharide antioxidant,free radical scavenging,inhibition of apoptosis and regulation of endoplasmic reticulum stress.First of all:Micro oxidative injury model of vascular endothelial cells induced by oxidized low density lipoprotein in human brain is established,and then analyze the model of HPS protection mechanism.
