摘要

Abstract

Objective To investigate the effect of microRNAs on the recovery of spinal cord injury in mice.Methods Target genes and miRNAs were searched and predicted,the results were confirmed in 293T cells.Spinal cord injury model was constructed in mice.The lentivirus-mediated miRNAs were injected via tail vein in mice with spinal cord injury.BBB behavioral experiment was performed and the morphology of spinal cord was observed.The expression of markers and target genes were detected.Results The miR-9,miR-124 and miR-125 induced the down-regulation of three or more target genes in vitro.miR-9 and miR-124 promoted the recovery of spinal cord injury caused by clamping;miR-9 group was better on movement function recovery,however,the effect of miR-124was worse than that of control group.In miR-9 group,GFAP was down-regulated,MAP2 and NeuN were up-regulated,and the target genes GLIS3 and CYBRD1 were also down-regulated.Conclusion MiR-9 may promote the repair of spinal cord injury by down-regulating the expression of target genes GLIS3 and CYBRD1 in mice with spinal cord injury.
