摘要

Abstract

Objective To explore the role of LKB1/AMPK signaling in the process of salen-Mn(salen-Mananese) inhibiting epithelialmesenchymal transition of bladder cancer cells.Methods MTT was used to detect the cell viability of bladder cancer T24 cells after salen-Mn treatment.Wound healing assay and Transwell assay were performed to detect the effect of salen-Mn on migration and invasion of bladder cancer cells.Western blot and real-time PCR were used to detect the protein and mRNA levels of E-cadherin,N-cadherin and Vimentin in bladder cancer T24 cells treated with salen-Mn.Western blot and siRNA transfection were pedormed to detect the function of LKB1/AMPK signaling in the salen-Mn-regulated epithehal-mesenchymal transition.Results Salen-Mn inhibited the proliferation,migration and invasion of human bladder cancer T24 cells(P < 0.05).Salen-Mn upregulated E-cadherin expression while downregulated N-cadherin and Vimentin in the mRNA and protein levels(P <0.05).And Western blot results revealed that salen-Mn upregulated the expression of p-LKB1 and p-AMPK.And the knockdown of LKB1 reversed high expression of E-cadherin,low expression of N-cadherin and Vimentin,and the inhibitory effect of migration and invasion induced by salen-Mn.Conclusion These findings suggest that salen-Mn could inhibit epithelial-mesenchymal transition of bladder cancer cells by upregulating LKB1/AMPK signaling pathway.
