Abstract

The present study was aimed to explore the evolutionary history and the potential biological functions of microRNA 125b (miR-125b).Pre-miR-125b sequences of various species were downloaded from the miRBase,multiple sequence alignments were performed using the ClustalW2 software,and the evolution tree of miR-125b was constructed.To comprehensively understand the potential biological function of miR-125b in chicken,the expression level of miR-125b was detected in 15 different tissues of Gushi chickens at five developmental stages by qRT-PCR.The target genes for miR-125b were predicted using TargetScan and miRDB,and were analyzed by GO (Gene Ontology) and KEGG pathway enrichment.Our results suggested that miR-125b was highly conserved among different kinds of animals.In the embryonic stage of the thalamus,miR-125b was gradually increased with the development of embryos,but decreased gradually after the shelling,and the expression level of the miR-125b was significantly raised.In the adult digestive organs (muscle stomach,glandular stomach,small intestine,and liver) miR-125b also exhibited a high expression level.Bioinformatics analysis showed that the predicted target genes of miR-125b were significantly enriched in the biological processes of tissue development,biological regulation,post-transcriptional regulation of gene expression,biological processing of small RNAs,extracellular matrix receptor interaction,and regulation of actin cytoskeleton,and in the MAPK signaling pathway,insulin signaling pathway,unsaturated fatty acid biosynthesis and NOD-like receptor signaling pathway.In conclusion,chicken miR-125b is ubiquitously expressed in all tissues examined and has temporal characteristic,and it may play an important role in the biological processes that are related to the development of many organs in hatched chicks.
