Abstract

Objective To investigate the effect of Huangqi Jiedu decoction on the expression of Jag1 gene and CyclinD1 protein in MCF-7 cells of human breast cancer and to reveal its possible mechanism of inhibiting the recurrence and metastasis of breast cancer. Methods Drug-containing serum of Huangqi Jiedu decoction group, doxorubicin group, Shenyi capsule group and normal saline serum group were built by using the method of gavage in SPF SD rats. The optimal concentration of cell inhibition routine culture of cells was detected by MTT. The breast cancer was intervened with 20% drug containing serum. The expression of JAG1 protein was detected by real-time quantitative PCR and CyclinD1 protein was determined by Western-blot. Results Compared with the normal saline group, the expression of Jag1 and CyclinD1 protein in the other serum groups significantly decreased (P<0.01). The inhibitory effect of the adriamycin serum group was the strongest (P<0.01). There was no significant difference between the serum group of Huangqi Jiedu decoction group and Shenyi capsule group (P> 0.05). Conclusion Huangqi Jiedu decoction could significantly prohibit the expression of Jag1 and CyclinD1, and provide the experimental evidence for Huangqi Jiedu Decoction in the treatment of breast cancer after surgery.
