载药电纺纤维膜防治双膦酸盐相关性颌骨坏死的研究
Experimental study on controlled release BMP-2 electrospun fiber membrane for prevention and treatment of bisphosphonate related osteonecrosis of jaws

Abstract
Objective:To detect whether the bone morphogenetic protein-2(BMP-2)membrane controlled release system could help prevention and treatment of bisphosphonate related osteonecrosis of jaw(BRONJ).Methods:A rat model of BRONJ was established by introperitoneal injection of zoledronate(ZOL)and tooth extraction,and evaluated by Micro-CT and histological examination.The poly lactic acid/polyethylene glycol(PLA/PEG)-BMP-2 membrane were preparedand the properties were tested.The BMP-2 membrane was implanted into the rat model to detect and analyze the mandibular speci-mens of rats.Results:Seventy percent of the rats showed typical BRONJ manifestations.The PLA/PEG-BMP-2 membrane could be stably degraded in vitro,continuously release BMP-2.Compared with the blank and control groups,the experimental group were better in bone healing,osteoclast function and soft tissue healing.Conclusion:In summary,local use the con-trolled release system of BMP-2 established by electrospinning technology in the bone trauma pit of the rat model with high risk of BRONJ could effectively reduce the incidence of osteonecrosis and improve the level of bone remodeling.关键词
