Mitochondrial structure and activities of mesenchymal stem cells derived from different human tissues

Abstract
Objective To compare the mitochondrial structure and activities of mesenchymal stem cells (MSCs) derived from different human tissues.Methods Mesenchymal stem cells were isolated from human adipose tissue,bone marrow,dental pulp and umbilical cord.The isolated MSCs were cultured and proliferated in vitro,and the purity was assayed by Flow Cytometry.The ultrastructure and metabolic activity of MSCs were detected by transmission electron microscopy and XFe Extracellular Flux Analyzers,respectively.Results MSCs derived from 4 tissues presented in culture as adherent growth and a spindle cell morphology,the cells positively expressed CD73,CD90 and CD105,and negatively expressed CD34,CD11b,CD19,CD45 and HLA-DR.Compared with MSCs from dental pulp and umbilical cord,the mitochondria of MSCs from adipose tissue and bone marrow had more mature morphology and higher metabolic activity.Conclusion Pluripotent stem cells derived from different tissues show similar characteristics,however,may have different ultrastructure and metabolic activity of mitochondria in MSCs of different origin.关键词
