摘要

Abstract

Objective To examine whether the MRL/lpr mice with autoimmune lacrimal gland disease can be used as a dry eye mouse model.Methods Thirty 8-week-old female MRL/lpr mice (n =15) and BLAB/C mice (n =15) were used as experimental group and the control group,respectively.The animals were observed at 8,12,16 and 20 weeks.Cotton thread test was performed and the tear volume was measured.H-E staining was performed for lacrimal gland tissues and the infiltration of lymphocytes was analyzed.The expression of TNF-α and IL-1β in lacrimal gland was detected by Western blot.Results Tear production was significantly decreased in the experimental group than that in the control group (P <0.05),especially at 16 and 20 weeks.HE staining showed more infiltrating lymphocytes in the experimental group in comparison with the control group.According to Western blot,the expression of inflammatory factors TNF-α and IL-1β in lacrimal gland was increased in experimental group (P < 0.05).With the progression of the disease in MRL/lpr mice,the infiltrating lymphocytes and the TNF-α and IL-1β levels increased gradually (P < 0.05).Conclusion MRL/Ipr mice present the similar manifestations of human dry eye and the physiological and pathological changes in lacrimal gland,indicating that MRL/lpr mice can be used as a stable and feasible dry eye model.
