Effect of overexpression of CD82 on OSCC-25 cell growth,invasion and apoptosis

Abstract
Objective To investigate the protein levels of CD82 and explore the effects of its overexpression on OSCC-25 cell prolifera-tion,invasion and apoptosis. Methods The eukaryotic expression vector pIRES2-EGFP-CD82 was transfected into OSCC-25 cells. The protein levels of CD82 were examined using Western blot at 0 h,24 h,48 h and 72 h after transfection. The non-transfected cells were chosen as blank group and the cells transfected with the NC plasmid group were chosen as negative control group. Cell prolifera-tion,invasion and apoptosis were assessed by MTT,Transwell and Annexin Ⅴ-FITC/PI staining,respectively. Results When the eukaryotic plasmid pIRES2-EGFP-CD82 was transfected at 72 h,the expression of CD82 protein was highly increased using Western blot analysis(P<0.05).Compared to blank group and negative group,the proliferation and invasion of OSCC-25 cells were significant-ly inhibited in CD82 transfection group,and the apoptotic rate of OSCC-25 cells was increased(all P<0.05). Conclusion The overexpression of CD82 may inhibit the proliferation and invasion of OSCC-25 cells,but enhance the apoptotic of OSCC-25 cells in vitro.关键词
