摘要

Abstract

To understand the evolution of PRRSV in Henan,a PRRSV strain was isolated in Marc-145 cells from the lung of suspected PRRSV-positive pig in Xinyang of Henan,and named as HNxy.HNxy strain could cause typically cytopathic effects.The results ofphylogenetic tree and homology analysis showed that HNxy belonged to the same cluster with high pathogenic PRRSV (HP-PRRSV)strains in China.The nucleotide homology of HNxy was the lowest with Lelystad virus,only up to 60.0%,and 89.2% with American classic strain VR-2332.The nucleotide homology of HNxy with Chinese HP-PRRSV strains JXwn06,JXA1 and HuN4 were 98.4%,98.3% and 98.4%,respectively.HNxy had the highest nucleotide homology with JXA1-P80(98.7%).The nucleotide homology of HNxy with NADC30 and NADC30-like strains HENAN-HEB,JL580,CHsx1401 were 84.6%,84.7%,87.3% and 83.6%,respectively.The results of sequence alignment showed that there were 30 discontinuous amino acid(aa) deletions in the high variable region of NSP2 and the epitopes of GP5 had some aa mutations.These results showed that HNxy belonged to a vaccine strain of highly pathogenic North American type PRRSV and had some mutations in the epitope region.
