摘要

Abstract

Macrophages RAW264.7 were stimulated and/or transformed with soluble schistosomula antigens(SSA) of Schistosoma japonicum and/or miR-181a mimics.The expression level of miR-181a,TNF-α and other cytokines,as well as M1 and M2 cell marker molecules were analyzed by real-time quantitative PCR or flow cytometry.The results showed that the expression of miR-181a,IL-1β,IL-4,TNF-α,IL-10 and IL-6 was up-regulated when the cells were stimulated with SSA.While IL-6 and IL-1β,TNF-α and INOS were down-regulated,IL-4 and arg-1 were up-regulated after RAW264.7 macrophages were transfected with miR-181a mimics alone.And the expression of IL-1β,IL-4,TNF-α,IL-10 and IL-6 was all down-regulated when the macrophages were first stimulated with SSA then transformed with miR-181a mimics.The results of flow cytometry showed that miR-181a mimics could down-regulate the expression of CD16/32,and up-regulate the expression of CD206.This study suggests that miR-181a may play a negative immune regulation role in macrophages stimulated with SSA,and promote the transformation of M1 to M2 macrophages.
