Protective effects of α-lipoic acid on ultrastructure damage by sub-chronic cadmium exposure in cerebral cortex of rats

摘要

Abstract

In order to investigate the protective effects of α-lipoic acid(α-LA) on ultrastructure damage by sub-chronic cadmium(Cd) exposure in cerebral cortex of rats,twenty-four 21-day-old female SD rats were divided into 4 groups randomly:control group,α-LA group,Cd-treated group,Cd and α-LA co-treated group.The rats of Cd-treated group were exposed to Cd(50 mg/L) in drinking water,and α-LA(50 mg/kg) was given to rats by gavage at the same time in the co-treated group.After 12 weeks,the mass concentration of Cd in cerebral cortex was measured by atomic absorption spectrophotometer,the changes in ultrastructure of the cerebral cortex were observed under transmission electron microscope and the expression of apoptosis related protein was measured by Western-blot.The results showed that compared with Cd group,α-LA could decrease the mass concentration of Cd in cerebral cortex significantly(P< 0.05),relieve the ultrastructural damage,and inhibit the decline of Bcl-2/Bax ratio and the increase of protein expression of cleaved caspase-9 and-3 caused by Cd significantly(P<0.05 or P<0.01).The results indicated that α-LA could protect the ultrastructure damage by sub-chronic cadmium exposure in cerebral cortex of rats.
