Antagonism Effects of Levosimendan on the Ototoxity of Gentamicin in Guinea Pigs

Abstract
Objective To investigate the protective effects of levosimendan and tetramethylpyrazine on hair cell injury induced by gentamicin in guinea pigs with deafness and its mechanism.Methods 60 normal Pregnant female DHP white guinea pigs were randomly divided into 6 groups:the gentamicin group,the ligustrazine positive control group,the hydroxypropyl-β-cyclodextrin adjuvant control group,the low dose levosimendan group,the high dose levosimendan group and the normal group.The auditory brainstem response (ABR) test was used to determine the levels of objective hearing threshold differences,and the levels of hearing thresholds were measured before and after modeling.The dyeing method and immunohistochemistry were used to observe the morphological characteristics of hair cells and the expression of apocytic protein Capase-3 and calcium channel protein CaV1.3 in different groups.Results The ABR threshold of gentamicin group was significantly higher than that of levosimendan drug protection group and ligustrazine positive control group (P=0.000<0.01;P=0.002<0.01),The expression of the gentamicin group of Caspase-3 protein and Cav 1.3 calcium channel protein in guinea pig cochlea was higher than that in low dose levosimendan group (P=0.004 <0.01;P=0.002<0.01).There was no significant difference in the dose of levosimendan group and gentamicin group.Compared with tetramethylpyrazine positive control group,Capase-3 protein was significantly higher than that of ligustrazine group (P<0.05),while that of Cav1.3 calcium channel protein was slightly lower than that of ligustrazine group differences (P>0.05).Conclusion The intravenous injection of levosimendan injection can significantly prevent the hearing loss of ototoxic guinea pigs.Under the action of low dose (1.5 mg · kg 1 · d-1) levosimendan,it may be through the inhibition of hair calcium Ion influx,to prevent intracellular calcium overload,significantly alleviate or delay gentamicin induced guinea pig cochlea hair cell apoptosis,in order to achieve the protection of the inner ear hair cell morphology and function of the role.关键词
