Abstract
Objective To investigate the inhibitory effect of sorafenib on non-small cell lung cancer H460 and A549 cell lines and its related mechanisms. Methods H460 and A549 cell lines were cultured and divided into four groups, the control group, 3.0 mmol·L-1, 6.0 mmol·L-1and 9.0mmol·L-1sorafenib group, MTT method was used to detect cell proliferation, cell cycle was detected by flow cytometry, Cyclin E1 and E2F1 protein expression were detected by Western blot. Results In different concentrations sorafenib group,the OD values of H460, A549 were significantly lower than control group (P<0.05), of which the OD value of H460 and A549 cellsin 9.0 mmol·L-1sorafenib group were (0.550±0.080) and (0.603±0.073), it was significantly lower than other groups (P<0.05); The G0/G1phase proportion of H460, A549 cellsin sorafenib groups were significantly higher than control group (P<0.05), the G0/G1phase proportion of H460, A549 cells in 9.0 mmol·L-1sorafenib group were (68.20±7.40)% and (69.12±7.04)%, which was significantly higher than other groups (P<0.05); Cyclin E1 and E2F1 protein expression in H460, A549 cells in different sorafenib concentrations group were significantly lower than those in control group (P<0.05), of which the Cyclin E1 and E2F1protein expression in H460 cells in 9.0 mmol·L-1sorafenib group was (0.206±0.097) and (0.412± 0.096), and they were (0.210±0.094) and (0.311±0.095) in A549 cells, which were significantly lower than other groups (P<0.05). Conclusion Sorafenib can inhibit the proliferation of non-small cell lung cancer H460 and A549 cells, and block the cells in the G1 phase, which may be related to the regulation of Cyclin E1 and E2F1 protein.关键词
