Abstract
Objective To assess the predictive value of VEGF and HIF-1α in colorectal cancer (CRC) patients with endostar resist-ance. Methods 260 CRC patients with combination of Endostar and chemotherapy drug in oncology department from February 2015 to June 2016 were collected, and they were divided into resistance group (85 cases) and sensitivity group (75 cases) according to anti-Endostar. Expression of serum VEGF and HIF-1α were detected using Elisa, and level of Survivin, LTEM1, and Hes1 mRNA, ASPP2, and PTEN mRNA was detected using real time PCR. Results The levels of serum VEGF (388.14±43.03) and HIF-1α (160.65±35.84) were higher in resistance group than in sensitivity group (P<0.001). The mRNA levels of Survivin, LTEM1, and Hes1 were higher in resistance group than in sensitivity group (P<0.001), while the mRNA levels of ASPP2 and PTEN were lower in resistance group than in sensitivity group (P<0.001). Logistic analysis revealed that the maximal contribution for prediction of anti-Endostar were VEGF and HIF-1α, their OR value were 7.352 (95% CI: 2.348~23.024, P=0.001) and 2.409 (95% CI: 1.166~4.978, P=0.018), respectively. Conclusion The expression of VEGF and HIF-1α were upregulated, and their levels could be used as the predictive function for endostar resistance.关键词
