Abstract
Objective:To investigate the effect of c-Jun N-terminal kinase (JNK) on the regulation of autophagy in the hippocampus of rats with subarachnoid hemorrhage (SAH) and the effect of edaravone.Methods:Male SD rats were divided into 4 groups randomly:sham operated (sham) group,SAH group,JNK inhibitor (SP600125) group and Edaravone group.The animal models were established with the two times blood injection in the classic pillow.SP600125 group was given lateral ventricle injection of SP600125 (3 μg/μl) in 30 min before modeling.Edaravone group was given Edaravone (10 mg/kg,1 time/24 h) through caudal vein injection after modeling operation had been achieved.The morphological changes of neurons in the hippocampus were detected with optical microscopy.The malondialdehyde (MDA) level in the brain was detected with thiobarbituric acid.The expression of phosphorylation JNK,Beclin-1 and LC3-Ⅱ of neurons in the hippocampus was detected with Western blotting and RT-PCR.Results:Compared with the sham group,the number of dead neurons,level of MDA and expression of phosphorylation JNK,Beclin-1 and LC3-Ⅱ of neurons in the hippocampus increased in the SAH group.Compared with the SAH group,number of dead neurons,level of MDA and expression of phosphorylation JNK decreased in the SP600125 group,but expression of Beclin-1 and LC3-Ⅱ of neurons in the hippocampus increased.Compared with the SAH group,number of dead neurons,level of MDA and expression of phosphorylation JNK decreased in the Edaravone group,but expression of P eclin-1 and LC3-Ⅱ of neurons in hippocampus increased.Compared with the SP600125 group,number of dead neurons and level of MDA decreased in the Edaravone group.Expression of phosphorylation JNK,Beclin-1 and LC3-Ⅱ I between SP600125 group and Edaravone group had no statistically significant difference.Conclusion:The activation of JNK can reduce the autophagy level of hippocampal neurons in SAH rats.Edaravone can inhibit the activation of JNK,improve the autophagy level of hippocampal neurons in SAH rats and reduce the neuronal injury in the hippocampus of SAH rats.关键词
