Effect of Nei Guan Electroacupuncture Preconditioning on Myocardial Ischemia Reperfusion Injury Model Rats

摘要

Abstract

Objective:To observe the effect of nei guan (PC 6) electroacupuncture (EA) preconditioning on cardiac myocytes protein kinase C(PKC),ATP-sensitive potassium channel(KATP) and mitochondrial permea-bility transition pore (MPTP) in myocardial ischemia reperfusion injury (MIRI) rats,and to explore the un-derlying mechanism for needling nei guan(PC 6) to prevent cardiovascular disease and the related meridian point zang-fu theory. Method:There were 40 male SD rats randomly divided into sham operation group , MIRI group,EA nei guan group and EA huan tiao group,10 rats in each group. Before establishing the MIRI animal model,EA nei guan group and EA huan tiao group were given EA 20 minutes per day,for 7 days. Ligation was conducted in the upper and middle 1/3 junction of the left anterior descending coronary artery to establish the animal model. After establishing the animal model ,electrocardiography was conducted. Ligating lasted for 40 minutes and was followed by reperfusion for 60 minutes. The heart tissue was sam-pled. The PKC,Kir 6.1 and MPTP were examined and the ultrastructure of cardiac myocytes was observed. Results:In MIRI group,PKC and Kir6.1 significantly decreased(P<0.01),while MPTP increased significantly (P<0.01),compared with the sham operation group. In EA nei guan group,PKC and Kir6.1 significantly in-creased(P<0.01),while MPTP significantly decreased(P<0.01),compared with MIRI group and EA huan tiao group. There was no significant difference between MIRI group and EA group(P>0.05). Conclusion:Nei guan EA preconditioning through activation of PKC,increases the opening of KATP,and inhibits the opening of MPTP,to relieve MIRI,and therefore it has a protective effect on the myocardium.
