摘要

Abstract

In order to evaluate the purification and extraction effects of different brands of SPE columns in pretreatment of pharmacokinetics experiments of Eprinomectin(EPR). Firstly,a certain amount of EPR standard solution was added to blank fetal bovine serum. Methanol was used to purify EPR in serum. Then the solution was extracted by Agilent's and Waters' C18SPE columns respectively,and was derivatized with N-methylimidazole and trifluoroacetic anhydride. The final products were determined by ultra performance liquid chromatography-fluorescence detector. At the same time,an equal amount of EPR standard solution was derivatized and determined by ultra performance liquid chromatography-fluorescence detector,and the result was taken as the control group. Besides,the absolute recovery rate was the criteria for evaluating the two brands of SPE columns. The results showed that,after being extracted by Agilent's and Waters'C18SPE columns respectively,the two EPR peaks were sharp symmetry without obvious impurity peaks,but there was a difference in absolute recovery rates,which were 94.50% and 85.79%,respectively. As a conclusion,the C18SPE column of Agilent company was more effective for EPR extraction.
