Abstract

Effect of topping on polyphenols metabolism, content of polyphenols and relevant key enzyme activities and gene expression using topping treatments on different periods (seedling stage and topping stage) in Honghuadajinyuan were tested by high performance liquid chromatography (HPLC), ultraviolet absorption spectrophotometry and quantitative reverse-transcription polymerase chain reaction (qRT-PCR). Compared with un-topped tobacco plants, topping at the 30th day after transplanting could lead to significant decrease of polyphenols such as chlorogenic acid in leaves, and the activity of PAL, PPO and POD increased, and 4CL activity decreased, while the activity of C4H was not significantly different from control.The expression of PAL1,PAL4 and 4CL2 were up-regulated.Topping at the 85th day after transplanting could significantly improve content of polyphenols in leaves, PAL, C4H, 4CL and POD activity increased, the activity of PPO decreased.The expression of PAL1 was down-regulated,and the expression of 4CL2 was not significantly different from control.Expression levels of PAL2,PAL3,C4H and 4CL1 were up-regulated in two periods.The above results showed that polyphenols biosynthesis in tobacco leaves induced by topping was related to growth period. Polyphenols content after topping was negatively correlated with activity of PPO, and positively correlated with activity of 4CL. At seedling stage the degradation rate of polyphenols in tobacco leaves was stronger than that in synthesis rate, resulting in the decrease of polyphenol content after topping. On the contrary, the synthesis rate of polyphenols in tobacco leaves was stronger than that in degradation rate after flower-bud appeared, which resulted in the increase of polyphenols content after topping.
