辨证运用骨蚀宁Ⅰ号、Ⅱ号对激素性股骨头坏死兔股骨头血管内皮生长因子表达的影响
Effects of Syndrome Differentiation-based Treatment with Gushining Ⅰ and Ⅱ on Expression of Vascular Endothelial Growth Factor in the Femoral Head of Rabbits with Steroid-induced Femoral Head Necrosis

Abstract
Objective To explore the mechanism of action of Gushining Ⅰ and Ⅱ in the treatment of steroid-induced femoral head necrosis,and to provide a theoretical basis for its clinical application.Methods Sixty rabbits were included in the study;6 were randomly selected as the normal group,and the other 54 rabbits were used to establish a model of steroid-induced femoral head necrosis.The model rabbits were randomly and equally divided into model group,positive control group,and treatment group.A triglyceride level of >1.00 mmol/L was considered as the presence of blood stasis syndrome.The model group was given normal saline by gavage;the positive control group was given Jiangusheng Pill by gavage;the treatment group received syndrome differentiation-based treatment with Gushining Ⅰ and Ⅱ (Ⅰ for blood stasis syndrome and Ⅱ for kidney deficiency syndrome).Six rabbits each were sacrificed at 4,8,and 12 weeks after model establishment in the model group,positive control group,and treatment group.The histomorphology of the femoral head was observed under a light microscope.The expression of vascular endothelial growth factor (VEGF) was measured by immunohistochemistry.Results Compared with the model group,the treatment group and positive control group had significantly alleviated osteonecrosis and a significantly lower percentage of vacant bony lacunae (P<0.05).At 4 weeks after model establishment,the model group had a significantly lower grey value of VEGF than the normal group (P<0.05),and the positive control group and treatment group had a significantly increased grey value compared with the model group (P<0.05).At 8 weeks after model establishment,the model group had a significantly higher grey value than the positive control group and treatment group (P<0.05).At 12 weeks after model establishment,the positive control group and treatment group had a significantly lower grey value of VEGF than the model group (P<0.05),and the treatment group had a significantly lower grey value of VEGF than the positive control group (P<0.05).Conclusion Gushining Ⅰ and Ⅱ can promote angiogenesis and osteonecrosis repair in steroid-induced femoral head necrosis by enhancing the expression of VEGF.关键词
