摘要

Abstract

In previous study,it was found that β-carotene was the key factor to improve laccase activity when co-culturing Pleurotus ferulae with Rhodotorula mucilaginosa.In order to reveal the specific mechanism,the full-length cDNA of laccase and its 5'-flanking sequence upstream of start codon were obtained and analyzed.QRT-PCR and 2-DE were used to work on laccase gene expression and the change of related proteins in the presence of β-carotene.A novel 1566 bp laccase full-length cDNA named LAC-CDS was cloned,containing the most conservative amino acid sequence L3 of laccase.Many putative stress responsive elements,such as XRE,STRE were identified,which was likely to involve in regulating laccase transcription.Laccase transcript level reached the peak in the presence of β-carotene after 5 days induction,causing 6 times of c,ontrol.Two-dimensional gel electrophoresis (2-DE) resulted in the identification of 9 different proteins,most of them related to protein synthesis and biological energy metabolism.The results indicated that β-carotene induced a high level transcription of laccase as well as influenced the synthesis of intracellular proteins.These would provide a theoretical basis for further enhancement of laccase fermentative level.
