Abstract

Lactococcus lactis exerts health benefits,inhibiting the growth of harmful bacteria,protecting the gut and enhancing human health.But in the practical application there is a big problem,which is difficult to preserve their activity and the activity is relatively low,especially in cryopreservation.In this paper,the perfect protective agent combination was determined by single factor and orthogonal experiment,including 30% trehalose,10% glycerol,20% sucrose,30% skimmed milk.Using this composite preservative agent,the survival rate of L.lactis after frozen could reach 91.96%.Furthermore,the effects of frozen cooling rate and frozen preservation after different thawing methods on the survival rate of L.lactis were investigated.The results revealed that the best cooling rate was-20 ℃/min and the best thawing temperature was 0 ℃.Under the above conditions,the survival rate was 93.14%.
