Abstract
Objective To observe the effects of ZNF268 gene interference fragment transfection on the proliferation, migration,and cell cycle of ovarian cancer SKOV-3 cells,and to explore the possible mechanism.Methods The SKOV-3 cells were divided into the observation group and the control group.The ovarian cancer cell line SKOV-3-sh-ZNF268 and the blank control cell line SKOV-3-sh-control were obtained by transfection of sh-ZNF268 and empty plasmids,respective-ly,in the observation group and the control group.MTT assay was used to observe the proliferation of the two groups(OD value);the soft agar colony formation test was used to observe the cell clone formation in the two groups(the clone forma-tion rate).Scratch test was used to observe the migration of cells in the two groups(the result was expressed as the number of migration cells).Propidium iodide single staining and flow cytometry were used to observe the cell cycle distribution. The expression levels of cyclin D2,cyclin E2,and CDK2 were detected by Western blotting.Results The OD values of the observation group at 1,2,3,4, and 5 days after transfection were higher than those of the control group(all P<0.05).The cell clone formation rate in the control group and the observation group was 9.7%±2.31% and 19.2 ± 1.45%,respectively;After 24-hour culture,the scratch was still evident,and the scratch width of the observation group and the control group was(2.53 ±0.12)um and(1.28 ±0.11)um; the number of migration cells were 67.59 ±6.54 and 136.92 ±10.28,respectively;significant differences were found in the above parameters between the observation group and control group(all P<0.05).The proportion of cells in the G0/G1,S,and G2/M phases was 62.3%±1.9%,23%±1.2%,and 14.7%±1.8%, respectively, in the observation group, versus 53.8%±1.8%,19.1%±0.4%, and 27.1%±2.2%,respectively,in the control group;compared with the control group,the proportion of cells in the G 0/G1 and S phases of the observation group increased,while the proportion of the cells in the G 2/M phase decreased(all P<0.05).The relative expression levels of cyclin D2, cyclin E2, and CDK2 proteins in the observation group were higher than those in the control group(all P<0.05).Conclusion Transfection of the transfected ZNF268 gene interference fragment could promote the proliferation of SKOV-3 cells,inhibit cell migration,and block the cell in G0/G1phase and S phase.Its mechanism may be realized by up-regulating the expression of cyclin D 2,cyclin E2,and CDK2.关键词
