青海云杉愈伤组织增殖与体胚成熟阶段影响因子研究
Study on the Influence Factors of the Proliferation and the Mature Stage of Somatic Embryo of Picea crassifolia
摘要
Abstract
In order to explore the influence factors of the proliferation and the mature stage of somatic embryo of Picea crassifolia, the mature embryo was used as explants, and the related research of somatic embryos was carried out. The results showed that: the best medium for the callus maintenance and proliferation stage of the Picea crassifolia was 1/2 LV + BA 1.1 mg/L+ 2,4-D 3.3 mg/L + 10g/Lsucrose + L-glutamine 400 mg/L + hydrolyzed casein 400 mg/L + Phytagel 3.5g/L. The best subculture cycle of embryogenic callus was about 14-16 days. During the development and maturation of somatic embryos, when the ABA concentration was 30 mg/L, the number of normal embryos obtained by two seed germinal callus was the most.关键词
