Abstract
Objective:To investigate expression of microRNA-17-5p( miR-17-5p) and its effect on myeloid cell leukemia-1(MCL-1) in pre-eclampsia(PE). Methods:40 preeclamp-sia pregnancies(mild PE 20 cases and severe PE 20 cases) and 20 healthy pregnant women (control group) were enrolled. Plasma and placental tissues were collected. Maternal plasma and placenta miR-17-5p, MCL-1 mRNA levels were detected by RT-qPCR. miR-17-5p and MCL-1 protein expressions in plasma and placenta were detected using immunohistochemistry. The correlation between plasma and placenta miR-17-5p,MCL-1 levels,clinical features and pregnancy outcome was also analyzed. Results:Compared to control group,the expression levels of maternal plasma and placenta miR-17-5p were significantly higher in PE group,MCL-1 mR-NA and protein levels were significantly lower in PE group(P<0. 05). There was no significant difference in plasma miR-17-5 p and MCL-1 mRNA between mile PE group and severe PE group (P>0. 05). The expression of miR-17-5p and MCL-1 mRNA in plasma,the expression of miR-17-5 p and MCL-1 mRNA、protein in placenta were all negatively correlated ( P<0 . 05 ) . The ex-pressions of plasma miR-17-5p and placenta miR-17-5p,plasma MCL-1 and placenta MCL-1 mRNA were all positively correlated ( P<0 . 05 ) . The plasma and placenta miR-17-5 p mRNA were positively correlated with systolic and diastolic blood pressure,while negatively correlated with gestational weeks of labor and neonatal birth weight ( P<0 . 05 ) . The plasma and placenta MCL-1 mRNA, placenta MCL-1 protein were negatively correlated with systolic and diastolic blood pressure, while positively correlated with gestational weeks of labor and neonatal birth weight ( P<0 . 05 ) . Conclusion:The expression of miR-17-5 p in pre-eclampsia increase in plas-ma and placenta,and it may be involved in the pathogenesis of PE by inhibiting the expression of MCL-1 .关键词
