低氧条件下人参皂苷Rg3对人喉鳞癌细胞分泌性VEGF的影响
The impact of VEGF in Hep-2 human laryngeal cancer cell induced by Rg3 in the condition of hypoxia

Abstract
Objective To investigate the effect of Rg3 to VEGF of secretion on human laryngeal carcinoma Hep 2 cells and its mechanism of inhibition to tumor angiogenesis.Methods Hep-2 cells culturing in hypoxia were divided into three groups:group Ⅰ (control group),group Ⅱ (Rg3 group),group Ⅲ (DDP group).Human vascular endothelial cells were divided into four groups:group Ⅰa (Rg3 group),group Ⅰb (DDP group),group Ⅱ (Rg3 supematant group),group Ⅲ(DDP supematant group).All of the cells were cultured in hypoxia for 48 hours.Collecting the supematant of Hep 2 cells of group Ⅱ and Ⅲ,which was used to culture HEVC of group Ⅱ and group Ⅲ.Immunocytochemical was used to detect the protein of HIF and VEGF.Detected the HIF and VEGF in the supematant of Hep 2 cells with ELISA.Then we take Mtt method to detect the inhibition of HVEC.Results Protein HIF and VEGF of group I in the supematant of Hep 2 cells was significantly higher than that of group Ⅱ and Ⅲ (F =15.627,P =0.023).There was no significantly different between group Ⅱ] and group Ⅲ.The HVEC inhibition of group Ⅱ was significantly higher than that of group Ⅲ (t =11.843,P =0.001).There was no significantly different between group Ⅰa and group Ⅰb.Conclusion The inhibition to tumor angiogenesis of Rg3 was stronger than traditional chemotherapy drug cisplatin in the condition of hypoxia.Rg3 inhibition of angiogenesis by at least the following two dimensions:first,inhibit the expression of VEGF;second,reduce the secretion of VEGF in promoting endothelial cell proliferation.But the effecting of reducing the secretion of VEGF in promoting endothelial cell proliferation worked by reducing the activity of VEGF itself or affecting endothelial function of VEGF receptor or some other ways to be further study.关键词
