Determine the content of total polyphenols in Anoectochilus roxburghii leaves by Folin-Ciocalteu colorimetry

Abstract
Objective To establish a Folin-Ciocalteu (FC) colorimetric method to determine the total polyphenols content of Anoectochilus roxburghii leaves.Methods The concentration of FC reagent and Na2CO3 reagent,reacting temperature and duration were evaluated respectively with gallic acid as reference and detecting wavelength 751 nm.The stability,precision and recovery rate of the method were also tested.Results The optimal conditions of FC colorimetry were:sample solution 1 mL,30% FC reagent 5 mL and 10% Na2CO3 10 mL,reacting temperature 25 ℃,reaction time 100 min.The calibration curve of polyphenols was well linear in the range of 0-50 μg · mL-1 (r =0.999 8) and the stability,precision and average recovery rate were 1.45%,1.18% and 3.79% by RSD (relative standard deviation).The average recovery rate was 101.10%.Conclusion The stability,precision and recovery of FC colorimetry were established well.The method is convenient and accurate,and it is reliable to be applied to determine total polyphenols content in Anoectochilus roxburghii leaf extracts.关键词
