摘要

Abstract

Objective To observe the effect of Ningmitai capsule on biofilm formation of Staphylococcus epidermidis,and to explore the possible anti-microbial mechanism of Ningmitai capsule.Methods Biofilm of Staphylococcus epidermidis was prepared in vitro.The effect of Ningmitai capsule on the biofilm-forming ability of Staphylococcus epidermidis was evaluated via 96-well cell culture microtiter plates with crystal violet staining as well as Congo red experiment.Then the change of tolerance to oxidative stress and sensibility to antibiotics for Staphylococcus epidermidis being treated by Ningmitai capsule was measured,qRT-PCR analysis was performed to show the relative transcript level of serp2195 and gpxA-2 upon Sepidermidis strain exposed to Ningmitai capsule at different concentrations.Results Ningmitai capsule significantly prevented biofilm formation in Staphylococcus epidermidis (P < 0.001) during crystal violet staining and Congo red experiment.Under the concentration of 20 mg/ml,Ningmitai capsule weakened the tolerance of Staphylococcus epidermidis towards oxidative stress for 8 fold.Similarly,Ningmitai capsule enhanced the sensibility of Staphylococcus epidermidis towards both chloramphenicol and erythromycin for 4 fold.Further study showed that Ningmitai capsule could downregulate the transcript level ofserp2195 andgpxA-2 to 22.9% and 24.5%,compared to the negative control respectively.Conclusion Due to the inhibition on biofilm formation,Ningmitai capsule can reduce bacterial drug resistance and work in synergy with other antibiotics in urinary tract infections and chronic prostatitis caused by biofilm-producing bacteria.
