Abstract

Objective:To explore the genetic diversity and genetic relationship among eight Polygonatum plants from the molecular level.Methods:Inter-simple sequence repeat(ISSR) was used to analyze the genomic DNA polymorphism of eight species of Polygonatum,and the genetic relationship was investigated by cluster analysis.Results:A total of 62 clear DNA marker bands were obtained from six primers,including 60 polymorphic bands and 96.8% polymorphic assay,which indicating that there was high genetic diversity among the eight species.The genetic similarity of the eight species varied from 0.4194 to 0.7258,with an average of 0.5904.The genetic similarity between Polygonatum nodosum and Polygonatum kingianum was 0.4194,and the genetic relationship was the farthest.The genetic similarity between Polygonatum nodosum and Polygonatum cyrtonema was 0.7258.In addition,UPGMA cluster analysis was carried out using NTSYS-pc 2.1 software.The results showed that eight species could be clustered into two groups with the similarity coefficient at 0.55.Conclusion:There is a low genetic similarity among the eight species of Polygonatum genus and there is significant genetic variation at DNA level,which has abundant genetic diversity.
