摘要

Abstract

Objective:To investigate the hypoglycemic effect of the extraction from Derris eriocarpa in vivo and in vitro.Methods:The glucose consumption(GC) and cell proliferation in high glucose model HepG2 cell culture medium were measured.Diabetic mice were induced by intravenous injection of Streptozotocin.Diabetic mice were randomly divided into model group,Metformin positive control group,and low-,middle-,high-dose (200,400,800 mg/kg)Derris eriocarpa groups,and normal control group were setup.Diabetic mice were intragastric (ig)corresponding drugs for 21 d.Fasting blood glucose (FBG)was measured weekly.After 21 d,pancreatic index,liver index,area under the curve (AUC),and the level of serum superoxide dismutase (SOD),malondialdehyde (MDA),glutathione peroxidase (GSH-Px) were measured.Pathological changes of pancreas were observed,immunohistochemical analysis was used to detected the expression of Caspase-3 and NF-κB in the pancreas tissues.Results:Hypoglycemic effect of petroleum ether extract in vitro was the optimum selection,the expression of FBG,MDA,AUC,Caspase-3,NF-κB of model mice were significantly decreased in vivo.The activity of SOD,GSH-Px in were increased(P<0.05 or P<0.01).The pathological changes of pancreas were improved.Conclusion:The petroleum ether extract of Derris eriocarpa has good hypoglycemic effect in vivo and in vitro.
