摘要
Abstract
[Objective]To study on extraction technology of the total sponin from the Agave sisaiana Perrine by enzymatic hydrolysis. [Meth-od] The extraction technology for the total saponin from the Agave sisaiana Perrine by methods of enzymatic hydrolysis-ethanol extraction,ur-solic acid as reference substance,the contents of total saponin from Agave sisaiana Perrine was determined by 5% Vanillin-Acetic acid-perchlo-ric acid colorimetric and spectrophotometer. The influence of enzyme concentration,enzymolysis reaction temperature,pH,enzymolysis reac-tion time and solid-liquid ratio were studied on extraction rate of the total saponin. [Result] The single factor experiment results showed that, enzyme concentration,enzymolysis reaction temperature,pH,enzymolysis reaction time had greater influence,but little influence by solid-liq-uid ratio. L9(34) orthogonal experiment results showed that, in the selected factors and levels, the influence degree of extraction rate was pH > enzymolysis reaction temperature > enzymolysis reaction time > enzyme concentration. The analysis variance showed that pH and tem-perature was most significant to extraction rate of the total saponin,and then the time. The optimum extraction technology were pH 5.5,enzy-molysis reaction temperature 45 ℃,enzymolysis reaction time 100 min,enzyme concentration 0.18 mg/mL,solid-liquid ratio was 1:20,The extraction rate was 19.20% in the condition. [Conclusion] The study provided reference for the total sponin from the Agave sisaiana Perrine by enzymatic hydrolysis.关键词
