摘要

Abstract

Objective To investigate time-dependentchanges in Leptin concentrations in brain tissue followingexperimental traumatic brain injury and to examine the relationshipwith cytokines such as IL-6 ,IL-1β,TNF-α.Methods The model of SD rat brain contusion was established byfree fall striking method ,and the random digital table was used to randomize the group .Rats weredivided into 3 groups based on thedepth of deformation :sham controls ,moderate injury and severe injury .Animalswere sacri-ficed on the 1st ,3rd and 5th days post-injury .The expression of IL-1β,IL-6 ,TNF-αand Leptin in different time was observed by enzyme-linked immunosorbentassay (ELISA) .Results One day after moderate injury ,IL-1β,IL-6 ,TNF-αand Leptin levelswere found to be markedly increased in the brain tissue .On the 3rd and 5th days ,the levels decreased to the shamcontrollevels .Follow-ing severe injury ,IL-1β,IL-6 andTNF-αlevels increased in correlation after the 1st day andreached the sham-control levels on the same days .However ,Leptin tissue levels decreased on the 1st and 3rd daysand normalized to the sham-control levels on the 5th day .Conclusion Leptin is decreasedin the early stage of severe injury .Leptin level correlates with degree of severetraumatic brain injury .
