Abstract
Objective:To study the effect of different concentrations of recombinant human growth hormone(rhGH) on the proliferation and osteogenic differentiation of bone marrow stromal cells cultured in vitro.Methods:Bone marrow stro-mal cells were isolated from rat bone marrow and cultured in vitro.Different concentrations of rhGH(0,2,5,25,50,150,500 μg/L)were added to observe the cell proliferation rate and detect the activity of intracellular alkaline phosphatase(ALP). Bone marrow stromal cells were cultured with osteogenesis induced in vitro,and the mRNA expressions of ALP,CoL-1 and OCN were detected by RT-PCR.Results:Different concentrations of rhGH could stimulate the growth of bone marrow stromal cells,enhance the activity of ALP and improve the expression of ALP,Coll,OCN mRNA in vitro.The mRNA expression of ALP,CoL-1,OCN,the activity of ALP and the proliferation rate was significantly increased at 150μg/L.Conclusion:Differ-ent concentrations of rhGH can potentiate the proliferation and differentiation of bone marrow stromal cells in vitro,and it has the most significant effect at the concentration of 150μg/L.关键词
