胡椒实时荧光定量PCR内参基因的筛选
The Identification of Internal Control Genes for Real-time PCR Normalization in Black Pepper

Abstract
In order to identify the suitable internal control gene for the gene expression analysis in black pepper,the expression of 9 housekeeping genes,including Polyubiquitin 1,Polyubiquitin 2,Polyubiquitin 3,GAPDH 1,GAPDH 2,Histone 3-1,Histone 3-2,Cyclophilin and Actin were assessed by using real time quantitative PCR in 8 different tissues.On the basis of Ct value comparison,the stability of housekeeping gene expression was analyzed by using geNorm,NormFinder and BestKeeper.The results of comprehensive ranking by three methods indicated that the most stable three reference genes were Histone 3-1,Polyubiquitin 1 and GAPDH 2.The value of pairwise variation Vn/n+1 suggested that Histone 3-1 and Polyubiquitin 1 had to be used together for normalization,which would make the expression of target genes more reliable in black pepper.关键词
