Abstract
Objective:To investigate the effect of MicroRNA-34a inhibits Notch1 combined with the research on the ability of invasion and proliferation of placenta tissues and human chorionic JEG-3 trophoblast cells. Methods:Preeclamptic placentas and healthy placentas (n = 30 each)were obtained after delivery from preeclamptic patients and healthy subjects.Chemically synthesized miR-34a mimic,miR-34a inhibitor and Notch1 siRNA were transfected into JEG-3 cells. The expressions of miR-34a miRNA and Notch1 mRNA were detected by Real-time PCR. The expression of Notch1 protein was detected by Western blot. The effect on the ability of invasion and migration was analyzed by Transwell migration assay. The effect on the ability of proliferation was detected by CCK-8. Results:The level of miR-34a was significantly elevated and Notch1 was significantly reduced in the preeclamptic placentas. Further,transfection with miR-34a mimic in JEG-3 cells inhibited cell invasion,migration,and proliferation.Transfection with miR-34a inhibitor in JEG-3 cells promoted cell invasion,migration,and proliferation.In addition,elevated expression of miR-34a reduced the expression of Notch1,inhibited expression of miR-34a increased the expression of Notch1. Downregulation of Notch1 expression significantly inhibited cell invasion,migration,and proliferation of JEG-3 cells. Conclusion: miR-34a may inhibits the invasion,migration and proliferation of trophoblast cells and play a role in inhibiting the expression of Notch1 contributing to the development of preeclampsia.关键词
