Abstract

Objective:To construct a new oncolytic adenovirus vector system,AD-hTERT-3E-EGFP-IRES-ELA. To observe its expression in ovarian cancer cell line SKOV3 and its effect on killing and inhibition of migration in vitro. Methods:Construction of adenovirus AD-hTERT-3E-EGFP-IRES-E1A by gene recombination. The expression of EGFP in SKOV3 cells was observed by laser confocal microscopy. The killing effect and mode of action of recom-binant adenovirus on SKOV3 cells were determined by MTS and flow cytometry. Cell scratches and Transwell technique was used to detect the effect of recombinant virus on the migration of ovarian cancer cells in vitro. Results:The oncolytic adenovirus AD-hTERT-3E-EGFP-IRES-E1A was successfully constructed. AD-hTERT-3E-EGFP-IRES-E1A expressed EGFP in ovarian cancer cells (SKOV3),and the fluorescence intensity increased with time. AD-hTERT-3E-EG-FP-IRES-E1A had a cytotoxic effect on ovarian cancer cells(P<0. 05)and inhibited the migra-tion of cells(P<0. 05). The main effect was the early apoptosis of SKOV3 cells. Conclusion:A novel tumor-specific adenovirus vector system,AD-hTERT-3E-EGFP-IRES-ELA,is successfully constructed. It can kill ovarian cancer cells and inhibit the role of tumor metastasis in vitro.
