多西环素对大鼠棘阿米巴角膜炎治疗作用的研究
Effect of doxycycline for the treatment of acanthamoeba keratitis in rats

Abstract
Objective:To investigate the effects and mechanism of antiamebic drugs combined with doxycycline in the treatment of acanthamoeba keratitis(AK) in rats.Methods:Thirty-two SD rats were randomly divided into a control group and an experimental group,with 16 cases in each group.All rats were used to establish AK model.The rats in the control group were given 0.02% chlorhexidine acetate eye drop combined with menstruum eye drop without doxycycline,while those in the experimental group were given 0.02% chlorhexidine acetate eye drop combined with 0.02% doxycycline eye drop.Slit-lamp microscope was performed to observe corneal ulceration,corneal infiltrate and corneal neovascularization.Inflammatory index of corneal disease was calculated.The corneal tissues were collected for histopathological analysis.The mRNA expression of matrix metalloproteinase-8 (MMP-8) was detected by quantitative real-time PCR.Results:Compared with control group,the experimental group showed slighter comeal ulceration,corneal infiltrate and corneal neovascularization.On day 3,5 and 7,the inflammatory index and the number of inflammatory cells in the experimental group were lower than those in the control group (P< 0.05),but no significant difference was noted between the two groups on day 10 (P>0.05).Furthermore,the expression of MMP-8 mRNA in the experimental group was down-regulated on day 3,5 and 10,compared with the control group (P<0.05).There was no significant difference in the expression of MMP-8 mRNA between the two groups on day 10 (P>0.05).Conclusion:Doxycycline could inhibit the inflammatory cell infiltration in AK rats,and the mechanism might be related to the down-regulation of MMP-8 expression.关键词
