Label-free与iTRAQ蛋白质组学方法在IgA肾病中的应用
Comparative analysis of label-free and iTRAQ approaches for urine proteomic analysis in IgA nephropathy

Abstract
Objective:To investigate the value of label-free and isobaric tag for relative and absolute quantification (iTRAQ) techniques application in IgA nephropathy (IgA-N).Methods:The morning urine were collected from 12 healthy controls and 12 patients with IgA-N.Proteomic analysis was performed to identify different expressed proteins.Results:Label-free mass spectrometry identified peptides length from 7-26,peaked in 10-18,and the maximum number was 800.iTRAQ identified peptides length from 6-26,peak distribution from 6-16,the maximum number was 1,400.The coefficient of variation (CV) within group was 5.7％-11.2％ calculated by iTRAQ,and was 3.7％-13.2％ by label-free.CV among groups was 7.9％-22.0％ (CV＜0.3) and 5.9％-19.0％ (CV ＜ 0.3) respectively by iTRAQ and label-free.Label-free identified 624 proteins,and iTRAQ identified 621 proteins.The volcano map analysis of label-free and iTRAQ could greatly screen different expressed proteins,and the clustering effect was good.GO analysis showed that Label-free and iTRAQ manifested high similarity of biological classification function.Conclusion:iTRAQ was greater than label-free in proteins identification especially in the low molecular weight.But CV from label-free was lower.The experimental scheme should be optimized according to the objective.关键词
