Expression of MMP-10 in non-small cell lung cancer tissues and its effect on proliferation and metastasis of lung cancer A549 cells

Abstract
Objective:To investigate the expression and clinical significance of matrix metalloproteinase-10 (MMP-10) in non-small cell lung cancer (NSCLC) and its effect on the proliferation and metastasis of lung cancer A549 cells.Methods:The expression of MMP-10 in NSCLC tissues and peficancerous tissues from 60 NSCLC patients was detected.The specific small RNA interference sequences (siRNA) for MMP-10 gene were synthesized and transfected into the A549 cells.The transcription of MMP-10 gene was detected by fluorescence quantitative PCR (qPCR) and the protein expression of MMP-10 was determined by western blotting.MTT assay was used to investigate the proliferation of the transfected A549 cells,and the cell migration was evaluated by transwell migration assay.Results:The protein expression of MMP-10 in lung cancer tissues was significantly higher than that in the peficancerous tissues (P＜ 0.05),and it was associated to lymph node metastasis (P＜0.05),but not correlated with patients' age and history of smoking (P＞0.05).MMP-10 mRNA and protein levels were significantly decreased after siRNA-MMP-10 transfection into A549 cells,which showed obvious reduction in the cell proliferation and migration (P＜0.05).Conclusion:The MMP-10 protein level in NSCLC tissues was significantly up-regulated,and was related to lymph node metastasis.Down-regulation of MMP-10 expression in lung cancer A549 cells could result in lowered cell growth rate and migration ability.关键词
