Spectrum-effect relationship analysis on stomach-nourishing effect of dendrobium officinale

Abstract
Objective To investigate the correlation between high performance liquid chromatography (HPLC) fingerprints and stomach-nourishing effect of dendrobium offcinale.Methods The water extract and ethanolic extract were made from dendrobium offcinale from 4 regions,and then HPLC was used to construct fingerprints. A hundred rats were divided into normal group,model group,water extract and ethanolic extract of dendrobium offcinale from each region group,with 10 rats in each group. Intragastric administration of distilled water was performed in the normal group and model group.Intragastric administration of corresponding extracts was performed in each dendrobium offcinale extract group. After the last administration,intragastric administration of alcohol solution was performed for gastric injury induction in all groups except the normal group.All rats were killed after 1 hour,then the gastric fluid was collected to detect the secretory volume of gastric fluid and pepsin activity,and the gastric injury was assessed.The partial least squares regression analysis was used to analyze the spectrum-effect relationship.Results The HPLC fingerprints of extracts from 8 dendrobium offcinale were established, and 30 characteristic peaks were confirmed.In the model group,the secretory volume of gastric fluid and pepsin activity were lower and the score of gastric injury was higher compared to the normal group(all P<0.05).Compared to the model group,the secretory volume of gastric fluid was higher and the score of gastric injury was lower in each water extract of dendrobium offcinale from each region group,the pepsin activity was higher in each water extract and ethanolic extract of dendrobium offcinale from each region group(all P<0.05).No.4, 8,15,18,21 and 23 chromatographic peaks were related to stomach-nourishing effect.Conclusion The HPLC fingerprints is closely related to the stomach-nourishing effect of dendrobium offcinale.The substances No.4,8,15,18,21 and 23 chromatographic peaks representing are probably the major active ingredients of dendrobium offcinale with stomach-nourishing effect.关键词
